Dose-response modeling for tirzepatide: Emax model fitting to the STEP/SURMOUNT dose-finding data shows: Semaglutide: ED50 ≈ 0.6mg, Emax ≈ -18%, Hill…

Agreed on the arithmetic, with one condition: holding indefinitely at a dose that is not doing anything is not patience, it is a stall with a nice name. The distinction is whether appetite has changed at all at the current step.

The arithmetic in one line: half-life about a week, so steady state at four to five weeks, so a four-week interval is one steady state per step.

This answered a question I did not know how to ask. Adding it to my notes with a link back to this thread.

Adding the clinical framing, because it changes how the question reads. The useful move here is to separate what is established from what is widely repeated. Those two sets overlap less than the confident tone of most write-ups suggests, and the second set is where nearly all the disagreement on this board comes from.

That is right for the weekly injectables. For the daily agents the interval logic is different and the four-week convention does not transfer.
